Role of cytoplasmic acetyltransferases, NAA60 and HAT1, in cellular protection against genotoxic agents

Histone acetyltransferases (HATs) are separated into two types. Type A HATs act on nucleosomal histones and thus primarily function in transcriptional regulation, while cytoplasmic (type B) known as enzymes that modify free before their assembly chromatin, may also outside the nucleus. N-alpha-acetyltransferase 60 (NAA60) is most recently discovered type B HATs, which N-terminal (NATs) found only multicellular eukaryotes. NAA60 localizes to Golgi complex possesses a unique ability catalyze acetylation of membrane-anchored proteins at N-terminus lysine side chains, biological significance remains unclear. To investigate cellular functions its relation other Hat1, we generated NAA60- or HAT1-deficient cells NAA60/HAT1-double deficient using chicken lymphocyte leukemia DT40 cell line. Although NAA60-deficient did not show any impairment growth showed slight sensitivity DNA damage agents, exhibited an additive increase methyl methanesulfonate (MMS) 4-nitroquinoline 1-oxide (4-NQO) when compared cells, were previously reported be moderately sensitive these agents.
